Oily skin is a problem for a large number of people, especially in tropical countries. This condition results in comedones, inflammatory acne, and other aesthetic problems in the skin. Emblica (Phyllanthus emblica L.) has a number of benefits for the skin; for instance, there were some studies that suggested that emblica has skin whitening effects, and anti-collagenase and anti-elastase activities; however, its anti-sebum efficacy has not been reported.The objective of this research was to study the anti-sebum efficacy of emblica toner on facial skin. The toner base was formulated, accelerated stability was tested, and preferences were evaluated in 10 volunteers. The toner base with the highest preference score was mixed with emblica extract. Then, the toner base and emblica toners were assessed for skin irritation by a single patch test in 30 volunteers. The anti-sebum efficacy was conducted using the randomized, single-blind, placebo-controlled, split-face method with unwashed and only-washed skin positions added to the middle of the forehead in the same group of volunteers assessed by a skin sebum measurement, SebumScale®, at 1 h before the test, once after washing, and 1, 2, 3 and 4 h after applying the toners on forehead and cheek skin. The stable toner base with the highest preference (85.6 ± 1.8%) was mixed with 1%, 2%, and 3% emblica extract. The toners were stable and did not cause any skin irritation. The 3% emblica toner was chosen for efficacy evaluation. The casual sebum levels of the forehead skin and cheek skin were 66.66 ± 7.01 and 56.12 ± 7.75 μg/cm2, respectively. The sebum level of the unwashed skin position changed (5.0 ± 1.66%) insignificantly up to 4 h (p > 0.05). In comparison, the sebum level of the only-washed skin position was recovered to the casual sebum level (99.4 ± 1.23%) within 3 h. Furthermore, the anti-sebum efficacy of the emblica toner (23.5 ± 1.24%) was higher than that of the toner base (12.0 ± 1.52%) (p < 0.05). The anti-sebum efficacy of emblica toner on cheek skin (26.9 ± 1.78%) was higher than that on forehead skin (20.1 ± 1.34%) (p < 0.05). In summary, the model of evaluation of anti-sebum efficacy used in this study has been found to be practical, and the emblica toner is safe and has apparent anti-sebum efficacy on facial skin....
Many personal care products, and particularly cosmetic products, contain preservatives that release formaldehyde. These are potentially harmful to consumer health, especially considering that the levels of formaldehyde in some products are hidden and excessive. Objectives: To study the formaldehyde levels of preservatives in personal care products and cosmetics on the UAE market and determine the extent of compliance with health and safety requirements. Methods and Materials: Sixty-nine personal care and cosmetic product samples from the UAE market were collected and prepared to determine their formaldehyde content. According to the Second European Commission Directive 82/434/EEC of 2000 and as per the Gulf Technical Regulation, Safety Requirements of Cosmetics and Personal Care Products in GSO 1943:2016, quantitative analyses were performed to identify and quantify the content of formaldehyde as free formaldehyde. Results: With a maximum permissible limit of ≤0.2% w/w, the average formaldehyde content was found to be 0.083 with a 95% CI (0.039–0.13). Nine of the tested personal care and cosmetic products exceeded the recommended formaldehyde level, corresponding to 13% of all samples. None of these samples listed the free formaldehyde content or formaldehyde releaser. Conclusion: Applying good manufacturing practices (GMP), education, and regulatory control to improve the regulation and inspection of cosmetics containing formaldehyde releasers as preservatives, conducting research, and reporting the adverse side effects are highly recommended. Several exopolymers with different chemical composition and correspondingly variety in their physico-chemical properties from halophilic microorganisms have still been described, however, with a low production yield. Chromohalobacter canadensis 28 isolated from Pomorie saltern synthesized an unusual exopolymer (EP) containing 72% γ-polyglutamic acid (PGA), an essential cosmeceutical additive. Current work suggests a novel approach for effective EP synthesis by C. canadensis 28 using continuous cultures. Highest production was observed at low dilution rates reaching a level of 2.1 mg/mL at D = 0.035, similar to those in batch cultures (2.34 mg/mL), however avoiding all disadvantages of discontinuous fermentation processes. At steady state, the total quantities of the synthesized EP after 48 h cultivation for the given equipment volume inD= 0.035 h−1 andD= 0.075 h−1 were 8.67 and 12 g, correspondingly, while it was 2.9 g for batch culture. Process parameters did not change after a ten-day run at D = 0.35 h−1. A degree of purity of EP fraction received from continuous cultures was significantly increased up to 93–96%. A lack of cytotoxicity and high cell viability were observed for human dermal fibroblast cells after 24 h incubation with crude EP from C. canadensis 28 and purified PGA fraction that could suggest its high potential for cosmetic applications....
Chemically synthesized retinyl palmitate has been widely used in the cosmetic and biotechnology industry. In this study, we aimed to demonstrate the microbial production of retinyl palmitate and the benefits of microbial retinyl palmitate in skin physiology. A heterologous retinyl palmitate biosynthesis pathway was reconstructed in metabolically engineered Escherichia coli using synthetic expression modules from Pantoea agglomerans, Salinibacter ruber, and Homo sapiens. High production of retinyl palmitate (69.96 ± 2.64 mg/L) was obtained using a fed-batch fermentation process. Moreover, application of purified microbial retinyl palmitate to human foreskin HS68 fibroblasts led to increased cellular retinoic acid-binding protein 2 (CRABP2) mRNA level [1.7-fold (p = 0.001) at 100 μg/mL], acceleration of cell proliferation, and enhancement of procollagen synthesis [111% (p < 0.05) at 100 μg/mL], strongly indicating an anti-ageing-related effect of this substance. These results would pave the way for large-scale production of retinyl palmitate in microbial systems and represent the first evidence for the application of microbial retinyl palmitate as a cosmeceutical....
It is currently estimated that one in every five Americans will develop skin cancer during their lifetime. Squamous cell carcinoma (SCC) is a common type of skin cancer that can develop due to the skin’s exposure to the sun. Herein, we prepared a topical gel containing 0.5% v/w phenethyl isothiocyanate (PEITC) for the treatment of SCC. PEITC is a naturally occurring isothiocyanate that has been shown to have efficacy against various types of cancer in preclinical studies. We first incorporated PEITC into a carbomer gel. A uniform formulation was prepared, and its viscosity was appropriate for topical application. We then demonstrated the release of PEITC from the gel into and through a Strat-M skin-like membrane. Finally, the effects of the PEITC-containing gel were tested against SCC and normal keratinocytes skin cells in culture, and these results were compared to those obtained for free 5-fluoruracil (5-FU), a commonly used skin-cancer drug. Our results show that a homogeneous PEITC-containing topical gel can be prepared and used to kill SCC cells. Thus, our formulation may be useful for treating SCC in the clinic....
